VeloGene Biotechnology Knockin Mouse Model Customizing Service

F0 "Founder" Ready - to -Use

Knock-in (KI) mice refer to genetically modified models in which exogenous sequences (such as reporter genes, tag sequences, mutation sites, or humanized fragments) are precisely inserted into specific genomic sites (typically endogenous gene sites) using gene-editing technology, enabling site-specific modification and functional studies of the target gene.

VeloGene’s TurboMice™ tetraploid complementation technology completes in situ KI editing at the embryonic stem cell (ESC) level, followed by aggregation with tetraploid embryos (which form only extraembryonic tissues such as the placenta) to compensate development.

  • In Situ ESC-Level Editing: Completes precise gene-editing at the embryonic stem cell level for high genetic fidelity.

  • Direct F0 Generation Homozygosity: All F0 generation mice originate from the same edited ESC monoclonal line, delivering homozygous and stably heritable KI models directly.

  • Rapid Delivery (3–5 Months): Achieves full generation and delivery within 3 to 5 months.

  • No Multi-Generation Breeding: Completely skips traditional multi-generation backcrossing and screening workflows.

Tradition Knockin vs TurboMice™ Knockin

MetricsTraditional ES Chimerism / Pronuclear Injection / CRISPRVeloGene TurboMice™ KI Customization
Time to Obtain Homozygous Usable KI Mice6–12 months (multi-generation intercrossing & screening)Delivery in 3–5 months
Long-Fragment / Multi-Component Knock-in (>10 kb)Low efficiency, prone to random insertion, unstable expressionESC homologous recombination, *in situ* precise integration
Same-Chromosome Multi-locus KI CombinationsLinkage drift, out-of-control breeding cyclesOne-time multi-locus knock-in via ESC monoclonal lines
F0 Direct Experimental UsabilityMostly chimeric / heterozygous, requires expansion validationYes, F0 mice are directly homozygous with full genotypes
Genetic Background ConsistencyProne to genetic drift through multi-generation intercrossingClone-level uniformity via same-strain ESCs
Cryopreservation & ExpansionSperm/embryo cryopreservation, recovery & breeding takes 1 year+ESC cryopreservation, batch delivery within 2 months

VeloGene Biotechnology's Knockin Mouse Solutions Advantages

VeloGene Biotech's TurboMice™ Knockin Service Can Cover Complex Scenarios

  • Reporter Gene Knock-in (e.g., Rosa26 or Col1a1 locus knock-in of EGFP/Luciferase/T2A-Cre)
  • Tag Protein Knock-in (e.g., 3×FLAG/HA/Myc fusion expression for protein interaction and localization studies)
  • Human Sequence / Functional Domain Knock-in (e.g., humanized antibody constant regions, immune checkpoint extracellular domain replacement)
  • Conditional Knockout (Flox) Mouse Generation (precise introduction of LoxP sites to prevent embryonic lethality)
  • Dual/Triple-Gene KI (e.g., multi-reporter gene co-expression, multi-target gene editing screening models)

VeloGene's Delivery Period(As quick as in 3 months)

  • Editing Strategy Customization: 1–2 days
  • Vector Construction: 2–4 weeks
  • ESC Editing & Monoclonal Validation (PCR/Sanger/WB): 3–6 weeks
  • Tetraploid Compensation Mouse Generation: 4–6 weeks
  • Weaning, QC, and Shipping: ~3 weeks
  • Homozygous Knockin Mice Delivered in as Fast as 3-5 Months (Rapid batch generation from ES cells to mice takes approximately 45 days at the fastest)

  • Order Now to Receive 2 Years of Free ESC Cryopreservation/Banking (No need for re-mating for subsequent cohort expansion; batch delivery in approximately 2 months)

Frequent Asks

Knockin mice from scratch traditionally take more than 6 months. Can you really deliver in 3 months?

Traditional method involves vector construction + ES chimerism + germline transmission + homozygous screening; whereas TurboMice™ completes the humanization all at once at the ESC level, with founder mice (F0) being directly homozygous, enabling the delivery of experiment-ready mice within 3 to 5 months.”

Affirmative. ESC homologous recombination is not limited by the DNA capacity constraints of pronuclear injection; VeloGene has delivered long-fragment KI orders exceeding 50 kb as well as multi-locus combination projects, ensuring site-specific integration and stable expression of exogenous sequences.

Positive. All somatic cells in F0 mice uniformly carry the homozygous KI genotype, and the expression of reporter genes/tag proteins is strictly validated by fluorescence detection/WB QC, allowing them to proceed directly to experiments such as cell tracing, protein interaction, and gene therapy evaluation without further expansion breeding.

Cryopreserved ESC clones are directly revived to edit lines, delivering live animals in batches within 2 months, featuring identical strain and origin with zero genetic drift.
